Paramecia culturing
We have three sets of two bottles, in total six bottles. One set is used for a week by taking 200mL of media for each day. On Mondays, you should bring the empty bottles from the biotron and make new media for two weeks later. This time, you should use 200mL of the media prepared two weeks before for the seed of the new cultivation, and use the old bottle for the feeding stock for the week.
Detailed protocol
- Wash two bottles that are brought from the biotron. Put three red beans, one Ebios tablet, and 900mL DW into each bottle, then sterilize them.
- After autoclaving, wait until the bottles return to room temperature.
- Fold the gauze twice and put it on the funnel on an empty glass bottle or beaker to purify Paramecia for cultivation.
- Select a set of bottles made two weeks before. For example, if you are making new media for set (3), the set to select should be set (1).
- Pour 200mL of Paramecia solution from the bottle for Mon/Tue/Wed into the new bottle through the gauze, and filter the debris out.
- Add filtered Paramecia solution into sterilized media.
- Incubate at the 28C incubator in 258.
